A Study of Tissue Culture and Rapid Propagation of Dendrocalamus brandisii Based on Embryo Sterile Seedlings

Abstract:Dendrocalamus brandisii (Munro) Kurz is an excellent species of giant woody sympodial bamboo for edible shoots and timber purposes, but the high price of seedlings from the conventional asexual propagation prevents it from the large-scale cultivation. To develop the tissue culture system of D. brandisii, the effects of plant growth regulators on rapid propagation and rooting in vitro are studied using the sterile seedling sprouting from seeds as explants. The results show that the optimal medium for axillary bud induction is MS+6-BA 3.0 mg·L-1+NAA 0.7 mg·L-1, with a 86.33% germination rate of axillary buds; the suitable medium for multiplication is MS+NAA 0.3 mg·L-1+6-BA 6 mg·L-1+TDZ 0.001 mg·L-1, and the multiplication coefficient can reach up to 4.90; the optimized medium for rooting is 1/2MS+NAA 2.0 mg·L-1+IBA 4.0 mg·L-1, with a 71.45% rooting rate. This study provides technical support for industrial seedling production of D. brandisii.
